Compensation and spreading: distinguish a tilted population from a wider one

Educational troubleshooting for laboratory professionals. Apply your validated local SOP; these examples do not establish clinical cut-offs or replace assay validation.

A bright CD8-positive population develops a broad cloud in the neighbouring detector. Increasing compensation until that cloud looks narrow can create an error: compensation addresses average spillover, while spreading reflects variability that remains after correction.

Systematic displacement

The secondary signal changes consistently with the primary signal. Recheck the single-stain reference, positive and negative populations, acquisition settings and fluorochrome identity.

Increasing width

The centre is reasonable but the distribution widens as the primary marker becomes brighter. Evaluate spillover spreading and panel design; a larger subtraction is not a remedy for lost resolution.

A practical diagnostic sequence

  1. Return to uncompensated single-colour data. Confirm the correct detector and that the positive reference is representative and sufficiently bright without saturation.
  2. Compare the negative reference with the sample preparation. Background differences can distort the estimate.
  3. Check that the reference uses the actual tandem conjugate and lot. Light exposure, storage or fixation can alter tandem behaviour.
  4. Inspect the fully stained panel with an appropriate fluorescence-minus-one control for the difficult boundary. An FMO shows the combined background and spread in that channel; it does not test binding of the omitted antibody.
  5. If separation remains poor, investigate fluorochrome assignment or titration under the validated panel-development workflow. Preserve the raw files and document changes.

Conventional versus spectral workflows

Do not apply a conventional compensation fix blindly to spectral unmixing. Spectral reference quality and autofluorescence modelling need their own review. A plausible-looking plot alone does not validate either calculation.
