Four-channel impedance cytometry separates overlapping events; clinical allergy testing is unvalidated
Summary
Lee and Tang demonstrated a four-channel, code-multiplexed impedance-cytometry proof of concept. Synthetic waveform decoding reached 99.85%, 96.93% and 88.83% for single, double and triple overlaps; these are not diagnostic accuracy figures. A six-donor basophil activation pilot remains preliminary.

Four-channel impedance cytometry separates overlapping events. Allergy diagnosis remains unvalidated.
Lee and Tang describe a code-multiplexed, multi-frequency impedance-cytometry system with four parallel channels and a deep-unfolding decoder. The paper is a peer-reviewed technical proof of concept in Microsystems & Nanoengineering (12:325, DOI 10.1038/s41378-026-01420-z), not a diagnostic product announcement.
What was measured
In a synthetic overlapping-signal benchmark, the algorithm identified the channel of origin for signals constructed from separately captured single-cell traces. Accuracy decreased as more waveforms overlapped: 99.85% for singlets, 96.93% for doublets, and 88.83% for triplets. This is a decoding benchmark, not the sensitivity, specificity or diagnostic accuracy of a basophil activation test.
| Signal overlap | Code-classification accuracy | Relative bar (0–100%) |
|---|---|---|
| Singlets | 99.85% | ████████████████████ |
| Doublets | 96.93% | ███████████████████░ |
| Triplets | 88.83% | ██████████████████░░ |
Biological test: basophil activation
Six donors contributed to a proof-of-concept experiment with four stimulation conditions. Electrical opacity was compared with CD63/CD203c fluorescence-flow measurements at the population level. These were not paired event-by-event measurements of the same basophils; the small donor sample and synthetic signal mixtures constrain interpretation.
What the technology changes
The technical bottleneck addressed here is not antigen detection but throughput while retaining event-channel identity. Label-free electrical phenotyping could complement fluorescence assays where membrane properties or activation states matter. Before clinical translation, a prospective allergy study would need predefined cutoffs, relevant patient controls, comparison to established basophil activation testing, intra/inter-run precision, interference and external-site reproducibility.
Commercial and regulatory status
Peer-reviewed proof of concept / WATCH. No commercial diagnostic availability, CE-IVD conformity or FDA clearance/approval is demonstrated in the source.

Claims Requiring Caution
Do not confuse decoding accuracy with diagnostic accuracy. Results for singlets, doublets and triplets concern classification of artificially overlapped electrical traces. They do not establish basophil activation assay sensitivity, specificity or external reproducibility. The six-donor experiment is preliminary; neither a commercially available medical diagnostic assay nor a CE-IVD/FDA authorisation has been demonstrated. Watch for larger donor cohorts, locked classification thresholds and paired clinical comparators.
Why It Matters
Code multiplexing could reduce the instrumentation required to acquire data from multiple microfluidic channels. The decisive next step is whether the signal-recovery advantage translates to reproducible, clinically interpretable measurements on natural overlapping events and diverse patient samples.
